It is with immense pleasure, we welcome you on behalf of the University of Agricultural Sciences, Dharwad and Indian Phytopathological Society (South Zone) to the National Symposium on "Plant Health Management a sustainable tool in addressing crop diseases under climate resilient Agriculture" scheduled to be held from 11-12 December, 2024 at the University of Agricultural Sciences, Dharwad. The symposium is organized under six theme areas to address the challenges being faced by the farming community, Crop Husbandry at national level with special reference to Southern zone. The symposium is also supported by various educational, entrepreneurial & media partners. The Integrated Disease Management Strategies under changing climate has become vital and is considered to play a key role for the future of sustainable and organic agriculture. This event is also focused on the growing significance of newly emerging pesticides and bioinoculants while, identifying newly emerging technologies in the league. In this Symposium, we bring together the academia, researchers, entrepreneurs, policy makers, progressive farmers and government officials comprising a core team of technical experts from across India to deliberate and understand the Plant Health Management technologies for use by farming communities across the country. The outputs would form a strong foundation in addressing the issues of global food security. SAMPLE ABSTRACT: An insight into Stenocarpella macrospora (Earle) B. Sutton causing Diplodia Leaf Streak (DLS) disease of maize through morphological and molecular characterization Prema G.U.1*, Pallavi K.N.2, Kruthika R.2,

Leaf Streak (DLS) disease of maize has been reported in Asia, Australia,
North America and Africa. The fungus that causes DLS is Stenocarpella
macrospora (Earle) B. Sutton, the synonym Diplodia
macrospora is also used. The first symptoms were noticed at Dharwad
on DKC 9133 maize hybrid during Kharif 2023. Studies on
morphological and molecular characterization of S. macrospora were
conducted at UAS, Dharwad during 2023-24. Symptoms first appeared as small
light tan to dark brown lesions on leaves. These lesions expanded into longer
lesions running parallel with the leaf. As the lesions elongate, concentric
zones were visible on the lesions. Leaf margins surrounding the lesion
appeared chlorotic and had a water-soaked appearance. As the disease
progressed on leaves, lesions merged to form large blighted areas. Typically,
symptoms were first observed in the lower to mid canopy; but, all leaves of
the plant were affected at later stages. Small black sphere-shaped fungal
structures called pycnidia were visible inside the mature lesions. Chains of
conidia bound together called cirrhi emerged from the ostiole of pycnidia.
The conidia of S. macrospora were 45 to 80 X 6 to 12 µm in
size, light tan to light brown, curved or irregular, and have one to two
septa. Another distinct characteristic of S.
macrospora was the white fuzzy mycelium when cultured on PDA.
Artificial inoculation was done by spraying conidial suspensions into the
leaf whorl of vegetative stage plants or placing dry infected material on
non-wounded leaves for pathogenicity studies. The plants sprayed with sterile
water were used as control. After fifteen days, typical leaf streak symptoms
were observed on the plants inoculated with S. macrospora.
Diseased leaves showed small brown lesions with a chlorotic halo. No symptoms
were found in the water-treated control plants. The seed-borne nature was
also confirmed by re-isolating the fungus from kernels of infected plants.
DNA from representative pure culture was extracted using the C-TAB method for
molecular identification. The ITS region was amplified with ITS1F/ITS4R
primers and sequenced. BLAST analyses of sequences of Dharwad isolate
of S. macrospora at NCBI database revealed >99.00%
homology with S. macrospora isolates and >96.00%
homology with S. maydis isolates. Even the Translation
Elongation Factor 1-alpha gene (TEF 1α) was amplified using TEF-F/TEF-R
primers, which showed >98.00% identity with S. macrospora isolates
and >85.00% identity with S. maydis isolates. Keywords: Stenocarpella macrospora, Diplodia Leaf Streak, Maize, Characterization |
The symposium will be held in Auditorium, College of
Community Science, Dharwad,
University of Agricultural Sciences, Dharwad.
